چکیده

H complexes of MutSand MutL-related proteins were identified from studies of DNA mismatch repair (MMR) and have been implicated in processing of recombination intermediates. In a recent issue of PNAS, Wang et al. (1) have reported several observations about the function of MutL-related genes of the yeast Saccharomyces cerevisiae in MMR and recombination in meiosis. First, they have observed that MLH1 is the common subunit in three different MutLrelated heterodimeric complexes, MLH1– PMS1, MLH1–MLH2, and MLH1– MLH3. Second, a possible role of MLH1– MLH2 in MMR was detected. Third, and most striking, MLH1–MLH3 was shown to play an important role in promoting crossing-over. This last observation extends the eukaryotic paradigm of combinatorial interactions between MutSand MutL-related heterodimeric complexes and mispaired bases to other types of DNA structures formed during replication, recombination, and repair.

mutS, mutL and mutH genes. A homodimer of MutS binds to mismatches in DNA and a homodimer of MutL coordinates mismatch recognition

sequences (recently reviewed in Buermeyer et al., 1999; Kolodner and Marsischky, 1999; Harfe and Jinks-Robertson, 2000; Hsieh, 2001). In the most thoroughly characterized Escherichia coli MMR system, which serves as a paradigm for eukaryotic MMR studies, initiation of MMR requires the products of mutS, mutL and mutH genes. A homodimer of MutS binds to mismatches in DNA and a homodimer of MutL c...

MutL traps MutS at a DNA mismatch.

DNA mismatch repair (MMR) identifies and corrects errors made during replication. In all organisms except those expressing MutH, interactions between a DNA mismatch, MutS, MutL, and the replication processivity factor (β-clamp or PCNA) activate the latent MutL endonuclease to nick the error-containing daughter strand. This nick provides an entry point for downstream repair proteins. Despite the...

MutS/MutL crystal structure reveals that the MutS sliding clamp loads MutL onto DNA

To avoid mutations in the genome, DNA replication is generally followed by DNA mismatch repair (MMR). MMR starts when a MutS homolog recognizes a mismatch and undergoes an ATP-dependent transformation to an elusive sliding clamp state. How this transient state promotes MutL homolog recruitment and activation of repair is unclear. Here we present a crystal structure of the MutS/MutL complex usin...

Stoichiometry of MutS and MutL at unrepaired mismatches in vivo suggests a mechanism of repair

Mismatch repair (MMR) is an evolutionarily conserved DNA repair system, which corrects mismatched bases arising during DNA replication. MutS recognizes and binds base pair mismatches, while the MutL protein interacts with MutS-mismatch complex and triggers MutH endonuclease activity at a distal-strand discrimination site on the DNA. The mechanism of communication between these two distal sites ...

The origins and early evolution of DNA mismatch repair genes—multiple horizontal gene transfers and co-evolution

To understand the evolutionary process of the DNA mismatch repair system, we conducted systematic phylogenetic analysis of its key components, the bacterial MutS and MutL genes and their eukaryotic homologs. Based on genome-wide homolog searches, we identified three new MutS subfamilies (MutS3-5) in addition to the previously studied MutS1 and MutS2 subfamilies. Detailed evolutionary analysis s...
